Il n&#8217;y a actuellement aucun traitement curatif et certains essais cliniques r\u00e9cents ont \u00e9chou\u00e9.<\/p>\n<p>L&#8217;ac\u00e9tylation des histones est un r\u00e9gulateur essentiel de la structure de la chromatine et de l\u2019expression des g\u00e8nes. Notre \u00e9tude d\u00e9montre un d\u00e9r\u00e8glement de l&#8217;ac\u00e9tylation de l\u2019histone H2B dans l\u2019hippocampe de souris d\u00e9veloppant des l\u00e9sions neurofibrillaires, l\u2019une des composantes pathologiques de la MA. Elle apporte en outre une preuve de concept qu\u2019un traitement in vivo avec une mol\u00e9cule activatrice de l&#8217;ac\u00e9tyltransf\u00e9rase CBP \/ p300 (CSP-TTK21) r\u00e9tablit les niveaux d\u2019ac\u00e9tylation de H2B dans l\u2019hippocampe et ainsi, restaure efficacement l&#8217;activit\u00e9 neuronale, la plasticit\u00e9 et la m\u00e9moire chez ce mod\u00e8le de souris.<\/p>\n<p>R\u00e9tablir une plasticit\u00e9 dans le cerveau malade en activant la fonction ac\u00e9tyltransf\u00e9rase de CBP\/p300 constituerait donc une approche th\u00e9rapeutique susceptible de retarder le d\u00e9clin cognitif et d&#8217;am\u00e9liorer les performances mn\u00e9siques chez les patients atteints de MA.<\/p>\n<p>&nbsp;<\/p>\n<p><strong>R\u00e9f\u00e9rence<\/strong><\/p>\n<p>Reinstating plasticity and memory in a tauopathy mouse model with an acetyltransferase activator.
