Au cours de notre \u00e9tude, nous avons identifi\u00e9 un nouveau syst\u00e8me de neuromodulation associ\u00e9 au r\u00e9cepteur m\u00e9tabotropique du glutamate mGlu4 dans l&#8217;amygdale, une r\u00e9gion particuli\u00e8re du cerveau impliqu\u00e9e dans la gestion de la douleur et des \u00e9motions. Avec nos collaborateurs, nous avons ensuite d\u00e9velopp\u00e9 et utilis\u00e9 l&#8217;optogluram, un ligand de petite taille, diffusible, dont la fonction est contr\u00f4l\u00e9e par la lumi\u00e8re. Ce nouveau ligand permet donc de prendre le contr\u00f4le de fa\u00e7on extr\u00eamement pr\u00e9cise et rapide des r\u00e9cepteurs natifs dans le cerveau d&#8217;un animal vigile et libre de ses mouvements. Gr\u00e2ce \u00e0 la lumi\u00e8re, nous avons d\u00e9montr\u00e9 la capacit\u00e9 du syst\u00e8me de neuromodulation et des circuits neuronaux associ\u00e9s \u00e0 mGlu4 dans l&#8217;amygdale \u00e0 r\u00e9guler rapidement et r\u00e9versiblement les diff\u00e9rents sympt\u00f4mes \u00e9motionnels (tel que l&#8217;anxi\u00e9t\u00e9) et sensoriels (hypersensibilit\u00e9 \u00e0 la douleur) associ\u00e9s \u00e0 la douleur persistante d&#8217;origine inflammatoire chez la souris. Ces r\u00e9sultats sont porteurs d&#8217;espoir du point de vue th\u00e9rapeutique car ils r\u00e9v\u00e8lent que le cerveau conserve sa capacit\u00e9 \u00e0 contrer les cons\u00e9quences de la douleur chronique et identifie une cible capable d&#8217;agir contre elle. En outre, il s&#8217;agit de la premi\u00e8re application de la photopharmacologie \u00e0 un mod\u00e8le animal pr\u00e9clinique pour r\u00e9v\u00e9ler la fonction de m\u00e9canisme de r\u00e9gulation endog\u00e8ne.<\/p>\n<p>Ce travail est le fruit d&#8217;une collaboration entre notre \u00e9quipe \u00e0 l&#8217;Institut de G\u00e9nomique Fonctionnelle \u00e0 Montpellier avec les \u00e9quipes de Chimie M\u00e9dicinale du Dr. Amadeu Llebaria \u00e0 Barcelone et du Dr. Francine Acher \u00e0 Paris, de l\u2019\u00e9quipe de Neuroanatomie du Professeur Francesco Ferragutti \u00e0 Innsbruck et de l\u2019\u00e9quipe de Neurophysiologie du Dr. Ingrid Ehrlisch \u00e0 T\u00fcbingen.<\/p>\n<p>&nbsp;<\/p>\n<p><em>Faced\u00a0with\u00a0the difficulty\u00a0of treating\u00a0chronic pain, we\u00a0questioned the central mechanisms\u00a0able to modulate the physical, affective and cognitive symptoms associated with it. In this study, we identify a new\u00a0neuromodulatory\u00a0system associated with\u00a0the\u00a0metabotropic glutamate receptors mGlu4\u00a0within the amygdala,\u00a0a key brain region linking pain sensation with negative emotions.\u00a0With our collaborators, we developed\u00a0optogluram, a small, diffusible ligand whose function is switchable by light. This new ligand allows to rapidly and precisely\u00a0photocontrol\u00a0native\u00a0receptors in the brain of freely moving mice. Using light, we demonstrate the capacity of mGlu4 receptors, and of the neurons expressing\u00a0them,\u00a0to rapidly and reversibly\u00a0abolish\u00a0sensory and affective symptoms of persistent pain such as hypersensitivity to pain, anxiety- and depression-related behaviors, and fear extinction impairment. These results are promising because they reveal that the brain conserves its capacity to counteract chronic pain consequences and identify a target against it.
