Ces travaux, r\u00e9alis\u00e9s chez la souris et publi\u00e9s dans la revue <em>The Journal of Clinical Investigation<\/em>, renforcent l\u2019id\u00e9e d\u2019un effet b\u00e9n\u00e9fique de la caf\u00e9ine sur les fonctions cognitives.<\/p>\n<p>La caf\u00e9ine est la substance psychoactive la plus consomm\u00e9e au monde. Alors que ses effets sur la vigilance et l\u2019attention sont largement connus, les m\u00e9canisme mol\u00e9culaires c\u00e9r\u00e9braux associ\u00e9s \u00e0 sa consommation r\u00e9guli\u00e8re demeuraient peu connus jusqu\u2019\u00e0 pr\u00e9sent. \u00a0Le Dr. David Blum du laboratoire \u00ab Alzheimer &amp; Tauopathies \u00bb du centre de recherche Lille Neuroscience &amp; Cognition (Inserm\/Universit\u00e9 Lille\/CHU de Lille) en collaboration avec le Dr. Anne-Laurence Boutillier du laboratoire de neurosciences cognitives et adaptatives (CNRS\/Universit\u00e9 de Strasbourg) vient de d\u00e9montrer, chez la souris, qu\u2019une consommation habituelle de caf\u00e9ine induit des changements mol\u00e9culaires durables dans l\u2019hippocampe, une structure essentielle \u00e0 la m\u00e9moire et qui d\u00e9g\u00e9n\u00e8re dans la maladie d\u2019Alzheimer. Les r\u00e9sultats montrent notamment que la caf\u00e9ine modifie de mani\u00e8re tr\u00e8s importante l\u2019\u00e9pig\u00e9nome hippocampique de mani\u00e8re diff\u00e9renci\u00e9e selon qu\u2019il s\u2019agisse des cellules neuronales ou non-neuronales. Cela concourrait \u00e0 favoriser le traitement de l\u2019information en r\u00e9ponse \u00e0 une tache d\u2019apprentissage, en exer\u00e7ant une action concert\u00e9e sur les cellules neuronales et non-neuronales.<\/p>\n<p>Cette \u00e9tude fait \u00e9cho aux travaux pr\u00e9c\u00e9dents du laboratoire Alzheimer &amp; Tauopathies sugg\u00e9rant un impact b\u00e9n\u00e9fique de cette mol\u00e9cule dans le contexte de la maladie d\u2019Alzheimer et \u00e0 l\u2019essai clinique de phase III en cours (<a href=\"https:\/\/www.cafca-alzheimer.fr\/\" target=\"_blank\" rel=\"noopener\">CAFCA<\/a>).<\/p>\n<p>&nbsp;<\/p>\n<p><em>English summary<\/em><\/p>\n<p><em>Caffeine is the most widely consumed psychoactive drug. However, there is a striking mismatch between the epidemiological evidence associating the regular intake of caffeine with benefits for chronic brain disorders and the molecular clarification of the impact of caffeine on brain function. Herein, using a combination of different non-hypothesis driven-omic approaches, we show that, in the bulk tissue analysis, chronic caffeine treatment reduced metabolic processes related to lipids, mitochondria and translation in the mouse hippocampus, some of which were identified at the different molecular levels analyzed, i.e. epigenome, transcriptome, proteome and metabolome. In sharp contrast to what was observed in bulk tissue, we found that caffeine induced a neuronal autonomous epigenomic response related to synaptic plasticity activation. These data were corroborated by the fact that caffeine treatment induced an increase in glutamatergic synapse proteins in the hippocampus and ultimately, enhanced transcriptomic regulations in response to learning. Overall, our data prompt the novel concept that regular caffeine intake promotes a more efficient ability of the brain to encode experience-related events.
